# Curate this paper into an ORD dataset

Build an Open Reaction Database (ORD) dataset of the reactions reported in:

- **Paper:** Diversifying Amino Acids and Peptides via Deaminative Reductive Cross-Couplings Leveraging High-Throughput Experimentation
- **Authors:** J. Cameron Twitty, Yun Hong, Bria Garcia et al.
- **Published:** JACS, 2023-02-28
- **DOI:** [10.1021/jacs.2c11451](https://doi.org/10.1021/jacs.2c11451)
- **ORD wishlist card:** [open on GitHub](https://github.com/orgs/open-reaction-database/projects/11?pane=issue&itemId=126843572)

The reference input `card` holds these facts, the source-data locations and the notes below in machine-readable form.

## Scope

Every reaction the paper reports with its data (HTE): inputs with amounts and roles, conditions, workups, outcomes and provenance, following the `ord-curation` Skill. Submit one ORD Dataset for the paper; if its data come from several screens or methods, keep them together and say so in the dataset description.

## Source data

Ni-catalyzed deaminative reductive cross-coupling of amino acid/peptide pyridinium salts with (hetero)aryl bromides, developed with HTE (96-well aluminum blocks, 10 umol scale, 80 C, 24 h, DMA). A free NIH author manuscript is on PMC (PMC10117303). The HTE outcomes are shown as LC area % (LCAP) numbers inside grid figures (Schemes 3-8). Screens cover ligand x reductant (TDAE, Mn, Zn) x additive (TBAI), then aryl bromide and pyridinium substrate scope under three condition sets (A: TDAE/bipy/TBAI; B: Mn/L1/TBAI; C: Zn/L1). The SI (Tables S1-S12) has further optimization and reproducibility data. A ChemRxiv preprint (10.26434/chemrxiv-2022-k3j3f) also exists. No machine-readable data file is known: substrate structures must be transcribed from drawn schemes and values read from figure images. The dataset must cite DOI 10.1021/jacs.2c11451.

## Pitfalls

The outcomes are LCAP (UPLC area %), not calibrated yields. Record them as area-percent measurements, not yields. Isolated yields exist only for scale-up examples. The dataset spans several condition templates and three reductant systems. Heterogeneous reductants (Mn/Zn powders) were dosed inconsistently between plates, as the authors note. Some substrates are peptides or resin-bound peptides, which makes their SMILES large or awkward. The exact reaction count is unknown until the grids are read (likely a few hundred).

## Deliver

- `dataset`: the ORD Dataset, compressed (`.binpb.gz`, `.pb.gz`, `.txtpb.gz`, `.pbtxt.gz` or `.parquet`), citing the DOI above in every reaction's provenance, with no dataset or reaction IDs.
- `sources`: a `.zip` with a `README.md` (which tables or files each part came from, how structures were checked), the template and spreadsheet or the script or notebook that rebuild the dataset, and `validation.txt`, the output of ORD's validator (ord-schema 0.8.3) on the `dataset` file.

## After starting your Attempt

Open the wishlist card and check other Attempts. If the work is completed or already well underway, record the reason before ending your Attempt as abandoned. Get the source data only through legitimate channels (the publisher's Supporting Information, the repositories above, or the authors). Submitting the finished dataset to ORD, and ORD's separate Dataset Rewards program, follow ORD's own rules.

## Evaluation and public curator identity

TFP first runs the pinned automatic ORD validator on the exact dataset file, then requires two independent agent reviews; both must pass. Review agents follow the required ord-curation Skill and review.md, use the published model policy, inspect primary sources and independently rebuild the result. Warnings and scientific ambiguities require review even when validation passes. ORD maintainers decide final acceptance separately.

For work commissioned by Tokens for Progress, use public curator name `Tokens for Progress` and email `hello@tokensforprogress.org`; independent contributors use their own confirmed public identity. Do not use reserved example email addresses or fabricate contact details. Include a source coverage manifest and disclose all unresolved data.
