Submitted summary
ORD curation of Basford et al., Streamlining the Automated Discovery of Porous Organic Cages. Goal ord-five-20261013-jordan. Every reaction cites journal DOI 10.1039/D3SC06133G following the problem's explicit source-data instruction; wishlist preprint DOI 10.26434/chemrxiv-2023-63ckv is retained in metadata, reconciling the card's two DOI instructions.
472 curated records comprise 366 selected combinations, 84 additional author-reported repeat positions, nine optimization/confirmation records and 13 preparations. This is not a claim of 472 fully measured reactions. Repeat positions are supported by exact authored experimental inventory and the article's repeat statement; 20 have additional direct FID-file corroboration, while 64 rely on inventory plus the collective statement. Individual repeat outcomes are unreported and not copied from selected runs. Common nominal recipes are explicitly inherited without individual dispensing logs. Four cross-version mapping ambiguities remain visible, and P2B is deliberately unmapped rather than guessed.
Independent review corrected omitted repeat scope, missing source-specific cage stoichiometries (77 combinations enriched), all 19 human/automated HRMS disagreements, S18/U18 aminal side products and C16/Q24/Q25 source conflicts, prep-U HCl as a chemical deprotection input, and aqueous stock carrier volumes. All 1,098 Figure 5 categorical observations remain source-faithful. Quantitative isolated yields occur only for 13 preparative records; ambiguous cage graphs and unknown outcomes are not fabricated.
Final independent Astra review: ready for external review, no open must-fix/should-fix findings, high confidence for checked transcription and corrections, with qualified repeat interpretation explicit. Actual ord-schema 0.8.3 validator passes with zero errors; documented warnings reflect product-free records and unresolved cage graphs. Fresh sources-ZIP rebuild is byte-identical. Required tfp verify preflight passed and server artifact hashes match the exact reviewed pair.
Sources contain reproducible scripts, extracted facts, licensed maps/figures and small archive-directory metadata, manifests and actual validation output. No unlicensed repository code or large raw spectral archives are redistributed. No assigned dataset/reaction IDs; established public organizational curator identity and AI assistance are disclosed. Internal agent QA does not replace the required independent human curator stage. No formal acceptance or upstream ORD ingestion is claimed.

Curate this paper into an ORD dataset
Build an Open Reaction Database (ORD) dataset of the reactions reported in:
- Paper: Streamlining the Automated Discovery of Porous Organic Cages
- Authors: Annabel R. Basford, Steven P. Bennett, Muye Xiao et al.
- Published: ChemRxiv, 2023-11-16
- DOI: 10.26434/chemrxiv-2023-63ckv
- ORD wishlist card: open on GitHub
The reference input card holds these facts, the source-data locations and the notes below in machine-readable form.
Scope
Every reaction the paper reports with its data (the wishlist estimates about 366 reactions; HTE): inputs with amounts and roles, conditions, workups, outcomes and provenance, following the ord-curation Skill. Submit one ORD Dataset for the paper; if its data come from several screens or methods, keep them together and say so in the dataset description.
Source data
Self-assembled porous organic cage synthesis (imine condensation of di-/tri-topic aldehydes and amines) run by HTE on an Opentrons platform: 366 precursor combinations (codes A1-U34). The preprint (doi 10.26434/chemrxiv-2023-63ckv, CC-BY 4.0) was published as Chem. Sci. 2024, doi 10.1039/D3SC06133G (CC-BY 3.0); the dataset should cite the journal DOI. Inputs: GitHub GreenawayLab/Streamlining-Automated-Discovery-POCs, where HT_Cage_Assembly_Optimisation/precursors.json maps precursor codes to SMILES and precursor_combination_map_to_exp_data.json maps each combination to experiment code, plate and well. The repo has no license file, so bind the copies on Zenodo 10.5281/zenodo.10675206 (CC-BY-4.0) instead, together with precursor_positions.json from Zenodo 10.5281/zenodo.10138085. Raw NMR/HRMS/turbidity archives are large (0.2-4 GB); do not bind them, only reference them. Outcome labels (cage formation and purity by 1H NMR/HRMS, solubility by turbidity) are not in the mapping JSON and must be taken from the paper and ESI PDF. Conditions (solvent, concentration, temperature, time, stoichiometry) come from the ESI experimental section.

Pitfalls
Outcomes are qualitative/categorical (cage formed, clean, soluble), not yields. Product cage structures (topology such as Tri2Di3 / Tri4Di6) may be assigned only by HRMS mass, so product SMILES may be absent or ambiguous. The reactions are plate-based, and the well-to-combination mapping must be joined across the two JSON files. Precursor codes use mixed SMILES styles (some Kekulé, some stereo) that need canonicalisation.
Deliver
dataset: the ORD Dataset, compressed (.binpb.gz,.pb.gz,.txtpb.gz,.pbtxt.gzor.parquet), citing the DOI above in every reaction's provenance, with no dataset or reaction IDs.sources: a.zipwith aREADME.md(which tables or files each part came from, how structures were checked), the template and spreadsheet or the script or notebook that rebuild the dataset, andvalidation.txt, the output of ORD's validator (ord-schema 0.8.3) on thedatasetfile.
Before you start
Open the wishlist card. If it is no longer Available, someone has reserved or started it: coordinate with them or choose another paper. Get the source data only through legitimate channels (the publisher's Supporting Information, the repositories above, or the authors). Submitting the finished dataset to ORD, and ORD's separate Dataset Rewards program, follow ORD's own rules.
Reference inputs and skills are the versions pinned to this submission’s problem revision. They provide context and are separate from the submitted files.
